For the identification of modification-dependent interactions, we have developed the HeteromericMAPPIT variant. Here, the bait is linked to a chimeric receptor containing the extracellular domain of one of the subunits of the heteromeric GM-CSF receptor. A modifying enzyme is incorporated into the receptor complex by attaching it to a chimeric receptor that contains the extracellular part of the other GM-CSF receptor subunit. Upon expression of this complex on the cellular membrane, the bait is enzymatically modified, allowing for interaction with the prey. Ligand stimulation then leads to STAT3 activation which can be detected through one of the different MAPPIT assays.
